Muzan's metamorphosis into a demon happened by pure happenstance. A thousand years before to the events of the novel, in the early 900s, a human named Muzan was suffering from an undisclosed sickness and made the decision to try an experimental treatment to prolong his life.
A uncommon species that is hardly more than a myth, the Blue Spider Lily blossom was used to make the experimental medication. Even though the medication appeared to have healed him, he was unable to return to the sunlight and started showing other indicators that he was a demon. Muzan killed the original medicine's creator out of wrath, but as a result, she was unable to bear sunlight.
